my Colts have great speed on defense. I play a man-zone cover 2 defense for the most part (4 D-Lineman, LB's/CB's cover man to man, 2 Safeties drop back into zone). My 4 lineman usually create a decent pass rush when the QB drops back, while I have a lot of man and deep zone coverage.
I find that it works against the run fine as well. Just make sure you hit *TRIANGLE, right (d-pad)* to shift your coverage guys directly in front of the player they're covering.
I change the scheme up a bit when playing against teams with different offenses (power running game, west coast offense, etc.), but my base defense is the one described earlier.
Watch what formation the other team is in. If they have 2 WR's in there, run a 4-3 formation. 3, a nickel formation. 4, a dime formation (unless your 4th CB really sucks...then leave it as nickel and let the LB or safety cover the 4th WR). If they're stacked (goal line), Run a 4-6 bear or goal line formation.
I hope that isn't too complicated...Justin pointed out the basics, I tried to get into a little more detail.
